ABI5 is a major regulator of late seed maturation and longevity acquisition in Medicago truncatula

Résumé

Abscisic acid insensitive 5 (ABI5) is a bZIP transcription factor that activates ABA- and stress-responsive genes in seeds and seedlings. In Medicago, we established that MtABI5 regulates the re-establishment of desiccation tolerance in germinated axes and certain LEA proteins during seed maturation. However, little is known about its role during seed development. A network-based approach (Verdier et al., Plant Physiol, 2013, 163:757–77413) suggests MtABI5 as a central regulator in the gene modules related to desiccation tolerance and longevity. Therefore, this study addressed the role of MtABI5 during seed maturation when longevity increases progressively over 30-fold. Characterization of T-DNA insertion mutants of Mtabi5 revealed that mutant seeds exhibited strongly reduced longevity and reduced accumulation of raffinose family oligosaccharides. To understand the underlying gene regulation related to the role of MtABI5 in late seed maturation, the transcriptome of wild type and Mtabi5 mutants was analyzed at different developmental stages. Comparison of these data with the transcriptome of ectopically expressed MtABI5 in Medicago hairy roots in the presence or absence of ABA revealed a restricted number of downstream regulated genes by MtABI5. Their role in final seed maturation will be presented and discussed in relation to seed longevity.
